Evolutionary Trace Annotation

The Evolutionary Trace Annotation (ETA) server suggests molecular functions for proteins structures based on local structural similarities to proteins of known function identified using 3D templates—six residue structural motifs of evolutionarily important amino acids chosen by the Evolutionary Trace.

Detailed instructions and examples are available, and clicking on the "i" beside each individual control shows help information specific to that control.

The various ETA components and code are also available for offline use upon request. Online and offline use of ETA is subject to these terms and conditions.

Enter a PDB ID

Enter the four character PDB code for the protein you wish to have annotated, plus a one character chain identifier if the structure contains multiple chains. If no Evolutionary Trace data is available to suggest a template for this protein, it will be generated automatically.

Upload a Trace

If you would like to use a customized Evolutionary Trace to generate a template, you may upload trace data in a zip archive, such as those generated by the ET Viewer.

Note: Make sure the custom protein submission for UET follows the PDB style id which is (1 digit + 3 letters + 1 letter for chain, use A for alpha fold structure) example : 5dddA

Suggested Residues
Custom Residues
Please select exactly six residues.

Submit Template

ETA suggests a six residue template based on available Evolutionary Trace data; you may also choose your own six residue template.
3. Amino Acid Labels

Add Custom Labels

Amino acid combinations allowed to match the template have been chosen based on a multiple sequence alignment for your protein; you may disable these suggestions or add your own, as well.
